Isometric is a standard 150, 90, 30 angle, A ping file of these three line could be created and used on multi-layers of a graphic program to create the building graphics. Then when done, erase the line layers.

Surfbored: Wow! But I agree with Kepli.
I was thinking something a little sharper for the angle, though. Almost directly down, but not quite. Just enough angle to give the impression of a single- or multi-story building, really. And with a size of a single 5-ft grid square equalling about 20 feet, it'ld be large enough to map out a town without losing too much detail.

I really don't think that iso maps are going to end up looking anywhere near "realistic", no matter how you do them. That said, I still think that working on them is a worthwhile endeavor.
And, I like Blu's idea. Pick a perspective and scale and go with it.
